Two Forms of CJC-1295:GHRH is produced in the hypothalamus, with its pulsatile release stimulating the pituitary gland to secrete GH. However, GHRH has a short half-life of just a few minutes. The first 29 amino acids of GHRH are responsible for its activity and are available as the synthetic peptide Sermorelin. To enhance its half-life to 30 minutes, Sermorelin is modified to create CJC-1295. The addition of a Drug Affinity Complex (DAC) further extends its half-life to eight days, resulting in CJC-1295 + DAC. Alternatively, CJC-1295 can be formulated without the DAC to mimic a natural GH spike each night.
The extended half-life of CJC-1295 means injections are only required once or twice weekly. However, the prolonged presence in the bloodstream provides a continuous stimulus for pituitary GH release, which is not a natural occurrence and may lessen GH pulse amplitude, potentially reducing GH stimulation in tissues.
Safety Considerations:When utilizing a long-acting CJC molecule, it is recommended to take 'hormone holidays' every three to six months for about three months to allow the pituitary gland to recover. During these breaks, Sermorelin is suggested instead of CJC-1295 + DAC.
These 'hormone holidays' could also minimize the risk of developing GH resistance, which can occur if antibodies form to inactivate GH or if the number of GH receptors decreases in tissues. Although these concerns are theoretical, no long-term studies have been conducted to fully explore them.
Potential side effects of CJC-1295 may include injection site reactions (such as irritation, redness, swelling, pain, or itching), headaches, diarrhea, flushing, temporary low blood pressure, nausea, and abdominal discomfort.
