CJC-1295, a powerful synthetic analogue of growth hormone-releasing hormone (GHRH), comprises 30 amino acids and is celebrated for its ability to enhance growth hormone and IGF-1 secretion while preserving the natural pulse of GH release.

There are two variations of CJC-1295:
The first is GHRH, which is naturally produced in the hypothalamus and triggers the pulsatile release of growth hormone from the pituitary, but its short half-life lasts only a few minutes. The active segment of GHRH comprises the first 29 amino acids, and it is available as a manufactured peptide known as Sermorelin. This was modified to extend its half-life to about 30 minutes, resulting in CJC-1295. Further enhancements, including the addition of DAC (Drug Affinity Complex), bind to the protein albumin, extending the half-life to approximately 8 days, known as CJC-1295 + DAC. There is also a non-DAC version of CJC-1295 that provides a more natural spike in growth hormone levels each night.
The long half-life of CJC-1295 allows for just one or two injections per week, providing a consistent stimulus for growth hormone release from the pituitary via the GHRH receptor. However, this may not replicate physiological conditions, potentially reducing the amplitude of GH pulses and decreasing stimulation of GH-responsive tissues.
Safety Considerations:
It is recommended to take 'hormone holidays' every three to six months while using long-acting forms of CJC. During these breaks, Sermorelin can be utilized as an alternative to CJC-1295 + DAC. These breaks can help mitigate the risk of developing GH resistance, which can occur due to the formation of antibodies that deactivate GH or a down-regulation of GH receptors in tissues. While these concerns are theoretical, long-term studies to confirm them are still lacking.
Possible side effects of CJC-1295 may include injection site reactions (such as irritation, redness, swelling, pain, or itching), headaches, diarrhea, flushing (vasodilation), transitory low blood pressure, nausea, and abdominal discomfort.
